Comenzando con las buenas noticias de la inducción positiva de ayer, vamos a comprobar la inducción con la construcción de nitrato con la proteína YELLOW. Continuando con las partes enviadas por el igem, Al a ver la PCR de colonia con cadmio y cobre solo se han obtenido tamaños adecuados de la construcción de cadmio, por lo tanto, con esta se han preparado 4 minipreps y stock de glicerol, para posteriormente sacar a placa y llevar a cabo las pruebas de inducción. Respecto al mercurio y al cobre, de los que no se han obtenido resultados positivos, se repitió la ligación y hoy se va a proceder a la electroporación de las partes. La construcción de arsénico en la que estaba pendiente de comprobar la presencia de RBS, se comprobó con el primer homólogo obteniendo resultados negativos, por tanto, se han hecho nuevas minipreps para hacer chequeo por digestión. 

Starting with yesterday's good news, we are going to check the induction with the nitrate + YELLOW construction. Continuating with the parts sent by Igem, checking the cadmium and cooper colony PCR we have only obtained satisfactory sizes with the cadmium construction, so, we have only prepared minipreps and glycerol stock with this last metal, as to plate them and start the induction test. Regarding mercury and cooper, from which we haven't obtained positive results, we have repeated the linking (leaving it overnight) and we are now going to proceed to the electroporation of the parts. The arsenic construction which was pending the verification of the RBS presence, has resulted to be wrong, so, we have done new minipreps to check them by digestion.

¡Ya estamos a juernes! Hoy, nuestro día en el laboratorio ha consistido en lo siguiente: en cuanto al cadmio, hemos hecho el preinóculo y hemos preparado soluciones de stock para posteriormente realizar la inducción. En relación al cobre y al mercurio, hemos hecho el checkeo por PCR y hemos picado a medio líquido. El arsénico es el que más guerra nos está dando: las colonias obtenidas hasta ahora eran erróneas, y hemos continuado checkeando para ver si había alguna colonia correcta. Y lo mejor de todo... Después de todo este tiempo, el trabajo con el plomo ya ha terminado. ¡Primer objetivo conseguido!

Evidentemente como cada día, al mismo tiempo que se han realizado procesos en el laboratorio, otros componentes del equipo se han estado ocupando de la wiki, el blog, y todos los demás procesos que del mismo modo tienen gran importancia en el proyecto.

It's almost friday (thursday)! Today, our day in the laboratory has consisted of the following: as regards cadmium, we have made the preinoculum and we have prepared stock solutions to subsequently make the induction. In relation to copper and mercury, we have done the PCR check and we have chopped in liquid medium. The arsenic is the one that is showing more resistance: the colonies obtained so far were wrong, and we have continued checking to see if there were any correct colony. And best of all ... After all this time, the work with lead is over. First goal achieved!

Obviously as every day, while  processes in the laboratory have been made, other components of the team have been dealing with the wiki, the blog, and all the other processes that are equally important in the project.
