Para terminar la semana, hemos realizado las PCR de colonia de la ligación del nitrato, ya con el blue y el terminador. Además, del arsénico, teníamos realizada la digestión con EcoR1 Spe1 y la hemos cargado en el gel para después purificar la banda. Y para seguir, hemos puesto la ligación con el plásmido que tiene la RBS para posteriormente, electroporar.

To finish the week, we have carried out the colony PCR of the nitrate ligation, already with the blue and the terminator. In addition, from the arsenic, we had digested with EcoR1 Spe1 and we have loaded it into the gel and then, we have purified the band. In the next step, we have put the ligation with the plasmid that has the RBS to subsequently electroporate.
