¡LA SEMANA LLEGA A SU FIN! // THE WEEK COMES TO ITS END!!

Ayer no fue un día muy novedoso: repetimos los procesos realizados en otras ocasiones (PCR con 3 tubos distintos), solo que a diferentes temperaturas, y uno de los tubos era el control, con el fin de ver que si el fallo de la primera vez estaba en la temperatura. Sin embargo, al ejecutar la electroforesis, pudimos comprobar que el error no residía en la temperatura ya que el resultado volvió a no ser el esperado. Pero traaaaaanquis, que de todo se aprende...¡ Roma no se hizo en un día!

Además de ello, comprobamos la eficiencia de nuestras bacterias a través de la fluorescencia que estas emitían a distintas concentraciones de nitrato, lo cual fue lo más satisfactorio de nuestra tard! ¡Mirad que chuliiiii!

Yesterday was not a very new day: we repeated the processes carried out on other occasions (PCR with 3 different tubes), but at different temperatures, and one of the tubes was the control, in order to see if the mistake of the first failure was the temperature. However, when performing the electrophoresis, the mistake was not found in the temperature, since the result was not the expected. But keep caaaaaalm… Rome was not built in one day!!!
In addition, we checked the efficiency of our bacteria through the fluorescence that is emitted from them at the different gathering of nitrate, which was the most satisfactory activity in our afternoon! Look at the result!

Y como nos gusta tanto perarar la PCR... repetimosssss! Hemos preparado más PCR, ¡nunca es suficiente! Y mientras la dejábamos reposar, nos hemos dedicado al checking por digestión de las minipreps de ayer. Para ello, hemos preparado un gel de agarosa y hemos introducido las enzimas de restricción a las minipreps, para más tarde ver el resultado y llegar a la conclusión de si las minipreps estaban bien o no.

Un rato más tarde, hemos ido a comprobar la PCR y... NOOOOOOOOOOOO! NO HA SALIDO BIEN (DE NUEVO). ¡Pero si algo somos es cabezones, así que seguiremos intentándolo!

And as we love preparing PCR... let's do it againnn!! We have prepared more PCR, it is never enough! And while we let it rest, we have dedicated ourselves to the checking by digestion of the minipreps of yesterday. To achieve this goal, we have prepared an agarose gel and we have introduced the restriction enzymes to the minipreps, in order to later see the result and to conclude if the minipreps were good or not.

After a while, we have gone to check the PCR and... NOOOOOOOOO! IT HAS NOT WORKED (AGAIN). But we are really really stubborn, and we will keep trying!
