¡Completamos la primera semana!

Hoy hemos comprobado la eficiencia de las células competentes contando colonias haciendo uso de la fórmula.

Tres horas después de la preparación de los minipreps y de la digestión con las enzimas de restricción hemos comprobado, a través de electroforesis, que las bacterias tienen la construcción que hemos introducido.

Para la electroforesis hemos mezclado los 20 μl de cada muestra con 3 microlitros de buffer. A continuación, introducimos en los pocillos el marcador y las muestras con el buffer. Conectamos los extremos del voltímetro y aplicamos 90 voltios. Tras 30 minutos el DNA se ha trasladado suficiente. Lo visualizamos y ¡¡ et voilà !! Hemos obtenido dos bandas, una que es la correspondiente a la construccion y otra correspondiente al resto del plasmido. 

Today we have checked the efficiency of the competent cells by counting colonies using the formula. Three hours after the preparation of the minipreps and the digestion with the restriction enzymes we have verified, through the electrophoresis, that the bacteria have the construction that we have said. For electrophoresis, we have mixed the 20 μl of each sample with 3 microliters of buffer. Next, we have introduced the marker and the samples with the buffer in the wells. We have connected the ends of the voltmeter and applied 90 volts. After 30 minutes the DNA has moved enough. We have visualized it and ¡¡et voilà !! We have obtained two bands, a relationship that corresponds to the construction and another part to the rest of the plasma.
