El 6 de agosto, hemos realizado la inducción con el nitrato (se han colocado distintas concentraciones de nitrato potásico), y estamos a la espera de los resultados.
Además, hemos checkeado las PCR del día anterior y hemos hecho el stock de glicerol con el arsénico. También hemos hecho la miniprep con el mercurio y el plomo (electroporando y dejando correr en el gel). En el caso del plomo, todas las colonias han resultado positivas, mientras que para el mercurio los resultados no han sido satisfactorios.
On August 6, we did the induction with nitrate (different concentrations of potassium nitrate have been placed), and we are waiting for the results.
In addition, we checked the PCR of the previous day and we made the stock of glycerol with arsenic. We also did the miniprep with mercury and lead (electroporating and letting it run on the gel). In the case of lead, all colonies were positive, while for mercury the results were not satisfactory.

Estos días, hemos avanzado tanto por la mañana como por la tarde.
A la mañana, hemos cogido las colonias existentes de arsénico y proteína amarilla y las de nitrato y proteñina azul y hemos hecho el inóculo.
El inóculo ha sido realizado dejando crecer la colonia y midiendo cada X tiempo la absorbancia de la misma comparándola con el medio de cultivo. Cuando se ha alcanzado el valor deseado (0.4) se han sometido las colonias a inducción.

These days, we have advanced both in the morning and in the afternoon.
In the morning, we have taken the existing colonies of arsenic and yellow protein and those of nitrate and blue protein and made the inoculum.
The inoculum has been made by letting the colony grow and measuring its absorbance every X time by comparing it with the culture medium. When the desired value (0.4) has been reached, the colonies have undergone induction.
