¡Y aquí estamos de nuevo!

Para empezar la semana, hemos vuelto a realizar PCR de mercurio.

Para ello, teníamos 14 colonias. Preparamos la mezcla y echamos 30 microlitros a cada eppendorf. Tras ello, fuimos a la cabina y en los tubos de LB, depositamos el antibiótico. Posteriormente, picamos las colonias y las mezclamos con lo que había en los eppendorf para finalmente, añadir la punta del resultado al antibiótico.

A continuación, de la construcción de nitrato realizamos la digestión para introducirle el promotor, ya que previamente no contaba con él, razón por la cual no obteníamos los resultados esperados.

Además, pusimos a crecer las bacterias del plomo (stock 16) por agotamiento de estrías y lo dejamos creciendo overnight.

¡Pero aquí no acabó el trabajo! Hicimos la PCR de mercurio para ver si las medidas eran correctas, y realizamos 2 electroforesis de arsénico.

To start the week, we have realized the mercury PCR.
For this, we had 14 colonies. We prepared the mixture and poured 30 microliters to each eppendorf. After that, we went to the cabin and in the LB tubes, we deposited the antibiotic. Subsequently, we chopped the colonies and mixed them with what was in the eppendorf to finally add the tip of the result to the antibiotic.
Then, from the nitrate construction, we carried out the digestion to introduce the promoter, since it did not previously have it, which is why we did not obtain the expected results.

In addition, we grew lead bacteria (stock 16) due to depletion of stretch marks and let it growing overnight.

But the work didn't end here! We did the mercury PCR to see if the measurements were correct, and we performed 2 arsenic electrophoresis.
