¡Hola de nuevo chicos! Para empezar la semana, no hemos tenido muy buenas noticias, ya que María, una de nuestras dirigentes, realizó una revisión del trabajo ejecutado durante la última semana y los resultados no han sido del todo satisfactorios: apenas había colonias o al checkearlas se ha comprobado que muchas no eran correctas. No tenemos claro dónde ha estado el fallo ya que hay muchos procesos a tener en cuenta, pero nada que no pueda rectificarse. Ya nos hemos puesto a repetir parte de los procesos y a avanzar en los que no tenían problemas, ¡lo conseguiremos!

Una vez puestos al día, nos pusimos a trabajar. Vayamos por partes: el nitrato únicamente sufrió electroporación; sin embargo, con los otros metales empleamos más tiempo. En primer lugar, repetimos la PCR del plomo, purificamos los trozos de ADN, realizamos la digestión y dejamos el proceso de ligación overnight. Esos mismos procesos fueron repetidos con el mercurio. Pasando al arsénico, también realizamos la electroporación y las dejamos creciendo en placas. Lamentablemente, el resultado volvió a ser negativo... ¡de todo se aprende!

The first day of the week, we have not had very good news, since Maria, one of our leaders, made a review of the work carried out during the last week and the results have not been entirely satisfactory: there were hardly any colonies or when checking them it has been verified that many were not correct. Once updated, work started. Let's explain what we did part by part: nitrate only suffered electroporation; however, with the other metals we spent more time. Turning to arsenic, we also executed electroporation and we left it growing in plates. Unfortunately, the result was negative again ... but we are learning little by little!
